In March of 2014, I will collect four, evenly spaced soil cores at each individual plant (10 cm depth), beginning at the main stem of the plant toward the shrub canopy dripline and in a straight line, using a clean auger. Extending along this transect, I will take an additional four soil cores in the interspace, each the same distance apart as the shrub canopy-type soil samples. All soil samples along the shrub-interspace gradient will be sampled from the north-facing side of the plant canopy as determined by a compass. In addition to the eight shrub-interspace samples, I will collect three additional soil subsamples (15 cm depth) under the shrub canopy and combine them into one bulk sample. I will not collect shrub-interspace samples for Ferocactus cylindraceus as this plant lacks a true canopy. Instead, I will collect two bulk samples: combining three soil subsamples (10 cm depth) in the rhizosphere and three in the interspace. I will also collect litter and plant tissue (leaves and twigs) from each individual plant. I will quantify the microbial community structure (i.e., fungi, AM mycorrhizal fungi, and bacteria) of the shrub-interspace soil samples using next-generation sequencing, specifically with Illumina technology.
