Trapping of Peromyscus californicus and P. maniculatus. We will sacrifice collected individuals and prepare tissue and museum specimens. We will be collecting at Upper and Lower Robertson Creek, Madrone Canyon, and Big Creek. DNA will be extracted from tissue and tail samples using a DNAeasy kit (Qiagen). In order to determine species relationships and verify sister-species pairs, a molecular phylogeny will be constructed from sequences from 2-3 individuals of each species of the mitochondrial genes ND3 and COIII and the nuclear genes Mc1r and LCAT. In addition, three egg and sperm protein binding pairs will be sequenced and various phylogenetic tests will be used to determine whether these proteins experience positive selection and whether there has been a change in rate of evolution in monogamous lineages.
