McLaughlin Reserve

I'd like to return to finish sampling Common Garter Snakes (Thamnophis sirtalis). I will collect a maximum of 20 samples in order to estimate population structure and gene flow with neighboring populations. Animals will be captured by hand from patches of suitable habitat on the reserve. I will also opportunistically sample tissue from road kill animals. I would like to return to McLaughlin to sample Rough-Skinned Newts (Taricha granulosa). A minimum sample of 20 individuals is required to estimate population structure and gene flow with neighboring populations. Newts will be captured by hand and dip net from patches of suitable habitat on the reserve. In murky, stagnant water where newts cannot be visually identified, I may also use small 18 x 10 minnow traps. A maximum of three minnow traps will be used at once and traps will be checked and emptied every three hours. For each newt, I will use a human skin biopsy punch (Acupunche Acuderm, Inc.) to remove a 5mm diameter skin punch from the dorsum (Hanifin et al. 2004). This procedure normally does not involve bleeding or other obvious trauma, and the skin punches heal quickly with regenerated skin. Skin punches will then be used for both the genetic analysis and to assay for TTX toxicity. Once I have collected tissue, each animal will be released back to the same location it was captured. I will also opportunistically sample tissue from road kill animals.
