Carpinteria Salt Marsh Reserve

During our previous trip to the marsh, we sampled areas of high, intermediate, and low salinity along main channels in the marsh. We also identified several microbial mats in areas that seemed to flood infrequently. We will sample these areas in this upcoming trip. Sampling in the channel will be done with a sediment corer to a depth of about 0.5 meters. We will collect samples from elevations that are always submerged and from areas that are submerged only at high tide. Triplicate samples will be collected at each elevation and in each area along the salinity gradient. Microbial mats will also be collected using sterile spatulas/tweezers and 50ml falcon tubes. Samples will be stored in a cooler on ice until transported back to the lab where we will subsample each core and mat sample. Microbial communities will be examined from these samples using molecular and cultivation based techniques. Salinity, pH, and temperature will be measured at each site prior to sample collection.
