We request permission to visit the Eastern Sierra Reserve and the Aquatic Research Lab to collect leaf material of Gentianopsis holopetala, the sierra gentian (Gentianaceae). This is part of ongoing studies in the systematics and phylogeography of several Gentianopsis species across North America. We are particularly interested in G. holopetala because of complex patterns of geographic diversity that we have already documented. Nuclear and chloroplast DNA sequence data show that this traditionally recognized species consists of two distinct genetic lineages, one in the southern Sierra Nevada and one disjunct in the San Bernardino Mountains and western Great Basin. The Valentine Reserve is located between known localities of these two lineages; therefore the populations of G. holopetala will be key in understanding the evolutionary relationships of these plants. As part of this visit, we request permission to sample one leaf per plant from 30 plants (if available) from two populations documented from the reserve, for genetic analyses at the University of Miami. In addition, we request permission to make voucher specimens from two individuals, one from each population, to ensure correct identification of the plants and for potential morphological analysis in the future. Depending on the accessibility of the populations, we expect to visit the reserve for 1-2 days. We will plan to find our own accomodations in the area.
