Boat sampling trips from the Richard Gump Research Station: transects from the open ocean beyond the fore reef into the back reef lagoon. The sampling line will match one of the LTER transects west of the entrance to the Cook Bay. Sampling trips will be conducted every day (weather permitting) during 3.5 weeks, with occasional frequent sampling over day-night cycles. We will carry out measurements of volatiles (sulfur compounds, isoprene, halomethanes), iodide and iodate, DMSP, N-osmolytes, optical and molecular characteristics of DOM, TOC, TON, POC, PON, TEPs, CSP, total surface irradiance, wind speed, air temperature, airborne ozone, underwater optics, vertical mixing, nutrients, chla, phyto- and bacterioplankton abundances and diversity (pyrosequencing of 16s for prokaryotes and 18s for eukaryotes). A few samples, including day and night, will be collected for metagenomes, metatranscriptomes and metaproteomes with the aim to study the differential expression of enzymes involved in the cycling of volatiles. Measurements will consist of sampling seawater volumes (few L) and bring them to the lab for analyses. In some instances, seawater filtration will be conducted on board the boat. In some other instances, larger volumes (tens of L) will be collected and incubated in the lab to assess, e.g., the effects of UV radiation and the microbial community on the production of volatiles. Underwater imaging of the reef will be used to characterize sampling sites. Organisms collected will be microorganisms: bacterioplankton, pytoplankton and microzooplankton (protozoans). No disturbances are expected to animals and plants or cultural resources. There will be no introduction of species or genotypes.
